My H1B extension was filed for regular processing last month. Can I now upgrade to premium processing, since the petition was originally filed before April 3rd?

No. As of April 3, 2017, the USCIS is not accepting any new premium processing requests for H1B petitions. This applies even if the petition was filed before April 3rd. The USCIS expects to lift the premium processing restriction by no later than October 2017. (17.Apr.2017)
